Atualmente isso \u00e9 poss\u00edvel utilizando o DNA ambiental ou eDNA (do ingl\u00eas environmental DNA). Esse eDNA consiste no DNA deixado por organismos atrav\u00e9s de fezes, muco, gametas, peles, carca\u00e7as, pelos, entre outros, em uma variedade de amostras ambientais, como solo, \u00e1gua ou at\u00e9 mesmo no ar<sup>1<\/sup>. Para identificar as esp\u00e9cies em uma amostra, as sequ\u00eancias de DNA registradas s\u00e3o comparadas a bibliotecas de DNA previamente estudadas e depositadas em banco de refer\u00eancia, como o GenBank<sup>3<\/sup>. Com a forma\u00e7\u00e3o e enchimento de seu reservat\u00f3rio, em 1982, a barreira natural para a migra\u00e7\u00e3o de peixes do trecho m\u00e9dio do rio Paran\u00e1 (queda de Sete Quedas) foi substitu\u00edda pela barreira artificial da barragem de Itaipu. Essa barreira artificial causou impactos nas assembl\u00e9ias de peixes adjacentes, como a redu\u00e7\u00e3o da atividade reprodutiva nos primeiros quil\u00f4metros a jusante da barragem<sup>7<\/sup>. Para permitir a migra\u00e7\u00e3o de peixes e mitigar o impacto ambiental da barragem, um sistema de passagem de peixes conhecido como Canal da Piracema foi criado em 2002, ligando o Rio Paran\u00e1 ao Reservat\u00f3rio de Itaipu<sup>8<\/sup>. Com esse m\u00e9todo registramos 123 esp\u00e9cies do qual\u00a0 37 foram identificadas com base em sequ\u00eancias dispon\u00edveis. Amostragem tradicional realizada em cinco\u00a0 anos,\u00a0 integrando\u00a0 quatro\u00a0 m\u00e9todos amostrais, registrou 136 esp\u00e9cies de peixes. Ou seja, em duas ocasi\u00f5es amostrais r\u00e1pidas registram os 90% das esp\u00e9cies registradas historicamente no canal. Tamb\u00e9m encontramos uma diminui\u00e7\u00e3o na diversidade de peixes de 2019 a 2020, com os dados de metabarcoding, provavelmente devido \u00e0 recente seca extrema experimentada no sudeste do Brasil. A maior diversidade de esp\u00e9cies foi registrada na boca do canal, no Rio Paran\u00e1, localizado em um vale protegido com a maior heterogenedade ambiental.
